hear many bros talking about 失焦. How can I know whether my 350D has problem or not?

I will try both the wide side and tele side.

but should I use manual focus or auto focus? or both?

失焦 happens in Auto Focus only.
Set the aperture to the largest (eg F1.4 or 2.8)and mount the camera on a tripod triggered the shutter by timer.

The object is ideally 3 dimensional with very strong contrast very close on both side to the subject at about 30-45 degree
